双波长法测定谷类和豆类作物籽粒中直链和支链淀粉的含量

摘要: [目的]采用双波长法同时测定谷类和豆类作物籽粒中直链和支链淀粉的含量.[方法]由直链淀粉-碘、支链淀粉-碘复合物的吸收光谱,确定直链和支链淀粉的波长对分别为495、565、530和654 nm,将其代入回归方程,可求出样品中直链和支链淀粉的含量.[结果]直链淀粉的浓度在 0~80 mg/L、支链淀粉的浓度在0~400 mg/L有很好的线性关系,即符合比耳定律.[结论]双波长法可以有效地消除吸收池和试剂等因素引起的误差,具有准确度高等特点,用该方法测定直链和支链淀粉的含量得到了很好的结果.
