摘要: [目的]提高玉米秸秆纤维素的降解率.[方法]从腐烂的秸秆、森林土及羊瘤胃液等富含纤维素分解菌的样品中筛选出降解纤维素的菌株.样品以玉米秸秆为碳源富集培养后,采用刚果红纤维素琼脂平板法初步筛选纤维素降解菌,再以CMCase(羧甲基纤维素酶)酶活性为指标进行复筛,对复筛获得的高效菌株进行组合培养,筛选出高效组合菌群,进行菌株鉴定.[结果]筛选获得了3株活性较高的纤维素分解菌,通过形态及16S rDNA序列分析对其进行种属鉴定N05、N13为枯草芽孢杆菌,N21为黑曲霉;并对其进行组合培养,得到1个较好组合NSS,其CMC酶活性为6.07 U/mL,比单菌株有一定程度提高.[结论]混合菌群的酶活优于单一菌株.
