植物内生贝莱斯芽孢杆菌AH216鉴定及其对玉米盐胁迫耐受性和生长的影响

Identification of endophytes Bacillus velezensis AH216 and its effects on salt stress tolerance and growth in maize

    目的 研究植物内生功能芽孢杆菌在盐胁迫下对玉米生长及土壤养分的影响。
    方法 本文选用一株分离自棉花(Gossypium hirsutum L.)内生环境的贝莱斯芽孢杆菌AH216,开展鉴定和IAA合成等促生相关功能性指标的测定,分别在非盐胁迫(普通土)和盐胁迫条件下进行玉米盆栽试验,分析接种菌株AH216对植物促生效果及对土壤养分的影响。
    结果 通过16S rRNA和全基因组测序,将菌株AH216鉴定为贝莱斯芽孢杆菌(Bacillus velezensis)。菌株AH216具有解钾、产蛋白酶和纤维素酶以及产IAA和嗜铁素的能力。在普通土壤和盐胁迫土壤中,接种菌株AH216后均显著促进玉米植株生长,总生物量、株高、茎粗、叶宽、叶面积和叶绿素提高的幅度为6.8%~108.3%。同时,接种菌株AH216能够改善土壤理化性质和提高酶活,使普通土壤中有效磷含量、速效钾含量、蔗糖酶和脲酶活性分别提高了15.4%、30.3%、44.2%和35.8%,而盐胁迫土壤则分别增加了18.8%和34.8%、60.4%和59.4%。
    结论 植物内生贝莱斯芽孢杆菌AH216菌株对玉米生长发育具有显著促进作用,能够提高土壤养分和土壤酶活水平,并提高玉米植株对盐胁迫的抗性。

     

    Abstract:
    Objective This study investigates the effects of a plant Bacillus velezensis AH216 on maize growth and soil nutrients under salt stress treatments.
    Methods Bacillus velezensis AH216, isolated from the endophytic environment of cotton (Gossypium hirsutum L.), was selected for identification and measurement of plant growth-promoting related functions such as indole-3-acetic acid (IAA) synthesis. Pot experiments with maize were conducted under both normal soil and saline stress soil conditions to analyze the effects of inoculation of with AH216 on plant growth promotion and soil nutrients.
    Results Strain AH216 was identified as Bacillus velezensis through 16S rRNA and whole-genome sequencing. The strain demonstrated the ability to solubilize potassium, produce protease and cellulase, as well as synthesize IAA and siderophore. In both normal and saline-stressed soils, inoculation with strain AH216 significantly promoted the growth of maize plants, with increases in total biomass, plant height, stem diameter, leaf width, leaf area, and chlorophyll content by 6.8% - 108.3%. Additionally, inoculation with strain AH216 improved soil physicochemical properties and enzyme activities, leading to increases in available phosphorus, soluble potassium, sucrase, and urease activity by 15.4%, 30.3%, 44.2%, and 35.8% in normal soil, and by 18.8% and 34.8%, 60.4%, and 59.4% in saline-stressed soil, respectively.
    Conclusion The plant endophytic Bacillus velezensis AH216 significantly promotes the growth and development of maize, improves soil nutrients, and soil enzyme activity, and enhances the salt stress resistance of maize plants.
