Objective The cloning and transcriptional activation analysis of the bHLH (basic/Helix-Loop-Helix) family gene GbbHLH130 from Sea-island cotton.
Methods The bioinformatics analysis and multiple sequence alignment were conducted for the GbbHLH130 protein, followed by the construction of a phylogenetic tree. Furthermore, the transcriptional activation of protein was experimentally validated using a yeast system.
Results The open reading frame (ORF) of the GbbHLH130 gene is 1020 bp; it encodes 339 amino acids, with a molecular weight of 38.01843 kDa and a theoretical isoelectric point of approximately 8.52. GbbHLH130 has a specific conserved domain, bHLH, at positions 273−323 of the amino acids. The phylogenetic relationship indicates that the GbbHLH130 protein is in the same branch as GrbHLH130 (XP_052477053.1) of Gossypium raimondii and AtbHLH130 (AT2G42280.1) of Arabidopsis thaliana. Analysis of the cis-acting elements of the promoter revealed that the GbbHLH130 promoter sequence contains elements related to light response, plant hormone regulation, and various stresses. The yeast two-hybrid system experiment indicated that this protein has transcriptional activation function.
Conclusion This study obtained a GbbHLH130 protein with transcriptional activation activity, providing certain references for exploring the stress resistance mechanism of Sea-island cotton.
